Body surfaces of reptiles as projection screens of evolution

The PhD thesis deals mainly with the coloration (and also scalation) of selected reptile species and explores evolutionary or ecological contexts indicated by these traits. The main topics can be roughly divided into two groups: 'antipredation signals' and 'effects of parthenogenesis'. The dominant method used in most studies is visual modelling. The work consists of the following case studies. 1) Blue-tongue skinks use their conspicuous blue tongues to threaten potential predators. We found that the tongue has a relatively high UV reflectance, a typical feature of intraspecific communication in lizards. Using visual models, we investigated how the blue tongue is perceived by the conspecifics and predators (birds of prey). In both visual models, the UV-blue tongue appears more conspicuous against the natural background than a pink tongue. In addition, in the conspecifics model, its hue partially overlaps with hues of UV-blue spots, which are sexually selected traits in various species of lizards. Thus, the UV-blue tongue seems to contribute to the effectiveness of the deimatic display and its possible role in intraspecific communication cannot be ruled out either. 2) We detected UV reflectance in non-pigmented areas of the skin in the Leopard gecko (Eublepharis macularius). Reproductive modes in large Branchiopoda (Crustacea) and their effect on genetic structure of populations

This bachelor's thesis focuses on the influence of the reproductive systems of four groups of so-called large branchiopods (Crustacea: Branchiopoda) on genetic variability in their populations. The groups include the orders of fairy shrimp (Anostraca), tadpole shrimp (Notostraca), clam shrimp (Spinicaudata), and conchostracans (Laevicaudata). This thesis primarily examines the populations in Europe. Reproductive strategies are one of the key factors that influence the genetic variability of populations. Other factors include historical events such as the breakup of the supercontinents Gondwana and Pangea and the last ice age, along with its associated processes (such as the colonization of new habitats after the end of the last ice age, etc.) The most genetically variable populations in Europe are gonochoristic (Branchipus schaefferi, Phallocryptus spinosa, Triops mauritanicus, Leptestheria dahalacensis, and Lynceus brachyurus), followed by populations with other reproductive systems. Based on current knowledge, some gonochoristic species and species utilizing a different reproductive system are divided into multiple genetic lineages. Reproductive strategy of the termite Silvestritermes minutus and its consequences for the life history and ecological succes

Mixed reproductive strategies are unique modes of reproduction, in which the organisms alternate sexual and asexual offspring production to benefit from both processes. In termites, such a strategy was first described in 2009 and dubbed Asexual Queen Succession (AQS). Reproduction in most termite species is based on a presence of one pair of long-lived reproductives, the primary king and queen, producing in a lifelong strict monogamy all other colony members using classical sexual processes. In a few rare species, the primary queen has been observed to be replaced at some stage of colony development by a harem of neotenic queens. These arise from unfertilized eggs via thelytokous parthenogenesis and reproduce with the founding king. While the workers, soldiers and winged dispersing reproductives remain to be produced sexually, new generations of parthenogenetic neotenic queens appear in the colony to supplement the harem. As long as the founding king is alive, the genetic constitution of most offspring remains identical as if the founding queen would still be alive, while the overall fecundity of the colony increases thanks to the large number of queens. Diversity and distribution of scorpions (Arahnida: Scorpiones)

This thesis is devoted to species diversity and biogeography of Scorpiones order. Bachelor work is presenting basic problems of this area and major split in higher classification of scorpions, which is still not firmly defined. In the work is also provided an overview of the current 15 families, to whom is the scorpions order divided by classification proposed by Soleglad & Fet (2003), including their distribution, the number of families and species they cover. Factors which had or still have an impact on the expansion of scorpions are also discussed. Described factors, which affects the distribution and diversity of scorpions, are: continental drift and Quaternary climate change (historical aspects) and temperature, parthenogenesis and human factor (environmental aspects). These factors influenced the biogeography of scorpions at different taxonomic levels, in different evolution time scale and on differently sized territories.
Mechanisms of asexual reproduction in reptiles

In the case of reptiles, there has been described an occurrence of an asexual reproduction, especially in the case of a group Squamata; the asexual reproduction of birds (Aves) has been observed, too. Multiple times the asexuality of reptiles has been originated. Occurrences of obligate and facultative asexual species have been described. Most obligate asexual species have theirs origin in one or more hybridization events between closely related species. However, within the family Xantusiidae two exceptions where asexuality apparently originated without hybridization were discovered. Facultative parthenogenesis was originally expected only among reptiles kept in captivity, where the females were separated from males for a long time. However, this assumption was later disproved. The majority of the specimens formed by facultative parthenogenesis has reduced viability, which can be caused by the imperfect mechanism of the formation of parthenogenetic offspring. The terminal fusion is considered to be the probable mechanism of creation of diploid oocytes in the case of facultative parthenogenesis reptiles. The cytological mechanism of the parthenogenetic offspring's genesis was described for the obligate parthenogenetic species just for genus Aspidoscelis. During oogenesis in this case is ploidy...

New case of mixed reproductive strategy and its adaptive significance in the neotropical termite Silvestritermes minutus (Termitidae: Syntermitinae)

Thanks to the progress in genetic methods in population ecology, many critical discoveries were recently made in the field of reproductive strategies of social insect. Among them is the description of mixed reproductive strategies, combining advantages of sexual reproduction with thelytokous parthenogenesis. The queens of such species produce sterile castes through classical sexual process from fertilized eggs, while future queens develop asexually from unfertilized eggs. This original breeding system was first described in several genera of ants, but it was found very recently, that it is not restricted to social Hymenoptera, since it has been identified also in the phylogenetically remote eusocial clade of termites. Switching between the sexual process and thelytokous parthenogenesis, now known as Asexual Queen Succession (AQS), which enables a continuity of genetically almost identical queen generations after the death of the founding primary queen, was first reported only in one genus of lower termites, i.e. Reticulitermes. Embryonic development of the unfertilized silkworm eggs

Transgenesis of silkworms has great potential for the development of silk with new properties as well as for the preparation of recombinant proteins for the use in biomedicine. Zabelina et al. (2015a) showed that transgenesis of parthenogenetic silkworms facilitates the selection and maintenance of transgenic homozygotes with stabile transgene insertions. However, the efficiency of transgenesis was less than 2 % compared to 60 % in the standard, non-parthenogenetic silkworms. The purpose of the present research was to explore the cause of this difference. Since transgenesis is normally performed at 25 °C but in the parthenogenetic silkworms at 15 °C (3 days incubation at this temperature is part of the protocol for the induction of parthenogenetic development), we assumed that the eggs incubated at 15 °C might have been injected with the DNA construct at unsuitable time. The work was therefore focused on the rate of embryogenesis at 15 °C in the eggs treated in different ways. Intensive cleavage of the control eggs (strain K23) was observed at 12 h after oviposition at 25 °C and between 24 and 36 h at 15 °C. The transgenesis of parthenogenetic silkworms is also complicated by the embryonic diapause. In the current work, diapause was suppressed by implanting PK1 ovaries into the non-parthenogenetic male hosts K23. Parthenogenetic development was activated by the heat shock in the chorionated eggs dissected from the implants. No cleavage was detected at 12 h after the acti-vation and nearly complete blastoderm was found at 48 h. In respect to the course of cleavage at 15 °C, transgene injection 24 h after the activating heat shock can be recommended. The eggs from endogenous ovaries of the K23 females, which also contained the implants of the PK1 ovaries, acquired partial capacity of parthenogenesis. Low rate of embryogenesis was also found in the transgenic clone VTG1. Current results suggest that more research is needed to understand and possibly explore differences in the rate of silkworms exposed to different treatments.
